The Effect of Sialic Acid on miR-320a and let-7e Expression in Human Glial Cell Line

Introduction: Sialic acid plays pivotal roles in various critical physiological events at molecular and cellular levels and also pathological processes. Changes in sialic acid concentration are observed in many pathological processes; for example, there are some available data on the evaluated level of sialic acid and neurodegenerative prevalence. It can be proposed that sialic acid can play a ...

Enzymatic synthesis of sialylation substrates powered by a novel polyphosphate kinase (PPK3).

Active inclusion bodies of polyphosphate kinase 3 and cytidine 5'-monophosphate kinase were combined with whole cells that co-express sialic acid aldolase and CMP-sialic acid synthetase. The biocatalytic mixture was used for the synthesis of CMP-sialic acid, which was then converted to 3'-sialyllactose by whole cells.

Production of N-acetyl-D-neuraminic acid using two sequential enzymes overexpressed as double-tagged fusion proteins

BACKGROUND Two sequential enzymes in the production of sialic acids, N-acetyl-D-glucosamine 2-epimerase (GlcNAc 2-epimerase) and N-acetyl-D-neuraminic acid aldolase (Neu5Ac aldolase), were overexpressed as double-tagged gene fusions. Both were tagged with glutathione S-transferase (GST) at the N-terminus, but at the C-terminus, one was tagged with five contiguous aspartate residues (5D), and th...

Specific Synthesis of Neurostatin and Gangliosides O-Acetylated in the Outer Sialic Acids Using a Sialate Transferase

Gangliosides are sialic acid containing glycosphingolipids, commonly found on the outer leaflet of the plasma membrane. O-acetylation of sialic acid hydroxyl groups is one of the most common modifications in gangliosides. Studies on the biological activity of O-acetylated gangliosides have been limited by their scarcity in nature. This comparatively small change in ganglioside structure causes ...

THE Jonma~ OF BIOLOGICAL CHEMISTRY

Enzymatic activity was measured by following both the disappearance of substrate, CMP-[14C]sialic acid, and the appearance of product, [14C]sialic acid, after separation by ion exchange column chromatography. The stoichiometry of this relationship was 1: 1. The activity of CMP-sialic acid hydrolase in crude homogenates of rat liver (in 0.25 M sucrose) was 53 units of enzyme per g of liver. The ...
